Bir tridekapeptid olan nörotensin ve merkezi sinir sisteminde bağlandığı reseptörlerin emosyonların oluşumu ve ifadesi ile ilgili yapılarda yoğun olarak yerleşik olduğu ortaya konmuştur. Nörotensinin dopaminerjik sistemlerle yakın ilişki içinde olduğunu bildiren çalışmaların yanısıra, nöroleptik ilaçlar ve nörotensinin yolaçtığı davranış değişiklikleri ile bunlara koşut oluşan dopaminerjik aktivite değişiklikleri arasında da benzerlikler olduğu gösterilmiştir. Nöroanatomik, nörofizyolojik ve farmakolojik çalışmalar ek olarak bazı şizofreni hastalarının beyin omurilik sıvısında nörotensin düzeyinin düşük bulunması, nörotensinin bir “endojen nöroleptik” olabileceği görüşünün ileri sürülmesine neden olmuştur. Biz bu yazıda, sözkonusu hipotezin ileri sürülmesine neden olan çalışmaların sonuçlarını gözden geçirerek, nörotensinin belirli dopaminerjik sistemlerin işlevi üzerindeki düzenleyici etkisine yönelik ipuçlarını aramayı amaçladık.

Neurotensin (a tridecapeptide) and its binding sites were shown to be primarily localized in the central nervous system structures presumed to be involved in the generation and expression of emotions. Neurotensin has also been shown to be closely related to the dopaminergic systems and there seem to be similarities between the behavioral changes, and related alterations in the activity of the dopaminergic system induced by the neuroleptic drugs and neurotensin. Data obtained from neuroanatomical, neurophysiological and pharmacological studies, and decreased concentration of neurotensin detected in CSF specimens obtained from some of the patients wtih schizophrenia led to the hypothesis that neurotensin may be an endogenous neuroleptic. In this article, we review the results of the relevant studies which led to this hypothesis, and attempt to look for clues to the presumed modulatory effect of neurotensin on the distinct dopaminergic systems.
