Given the wealth of data supporting the noradrenaline hypothesis of depression, there remains a need for a potent and selective noradrenaline reuptake inhibitor. Reboxetine is a novel, potent, and selective noradrenaline reuptake inhibitor developed as an antidepressant. Reboxetine phannacokinetics is linear and multipl-dose range up to 12 mg/day. The recommended clinical dose is 8-10 mg/day in two divided doses. The elimination half-life is approximately 12 h. In spite of its marked effects on the noradrenergic system, reboxetine is well tolerated. Reboxetine has high potency in inhibiting the noradrenaline uptake whilst having little effect on the uptake of serotonin. Furthermore, it has very low affinity for a wide range ofneuroreceptors such as dopamine or acetylcholine. Given the pharmacological profile, reboxetine is expected to be a selective and potent tool for both psychopharmacological research and treatment of depression.
