Hastalıklardan sorumlu olan genlerin saptanması işlemine gen izolasyonu, hastalık geni fiziksel haritalaması gibi isimler verilir. Bu işlem 1- gen haritalama sonrasında saptanan kritik bölgenin daraltılması, 2- bölgede yerleşik olan genlere yönelik sistematik mutasyon analizleri ve 3- saptanan DNA değişikliklerinin polimorfizim olmayıp gerçekten hastalıktan sorumlu mutasyon olduğunun ispatlanması aşamalarını içerir. Bölgenin daraltılması çalışmalarında bağlantı ve ilişkilendirme analizleri kullanılır. Mutasyon analizleri doğrudan ya da dolaylı olarak hasta bireylerde DNA değişikliğini gösteren yöntemlerdir. SSCP, DGGE gibi analizler dolaylı olarak mutasyon varlığını gösteren analizler iken DNA dizi analizi doğrudan değişikliğin tipini göstermemize yol açan en etkin yöntemdir. Hasta bireylerde mutasyon saptandığı zaman aynı etnik kökenden çok sayıda kontrol örneği çalışılarak bulunan gen değişikliğinin polimorfizim olmadığının ispatlanması gerekir.

Identification of genes causing human disorders is usually referred to as disease gene tracking or physical mapping of human disorders. This procedure includes 1- narrowing down the initial localization of the disease-related critical region (fine mapping), 2-identification of candidate genes residing in this particular region and subsequent application of systematic mutation detection assays and 3- confirmation of the causative relationship between mutation and disease. Both linkage and association methods can be used for fine mapping of the region. Presence of functional and locational candidate genes in the region must be assesed using various databases generated by the human genome project. Direct (DNA sequencing) and indirect (SSCP, DGGE etc..) mutation detection methods are further applied to identify any genetic variation in these candidate genes. DNA sequencing analysis is the most efficient way to demonstrate DNA variations directly. Once a nucleotide change is observed, a number of normal individuals from the same ethnic background should be tested to prove the causative nature of this particular variation.
