Depresyonun etyolojisini aydınlatmaya yönelik çalışmalar artarak devam etmekle birlikte, bugün elimizde olan veriler bu ruhsal hastalığın klinik ve etyolojik heterojenitesine bağlı olarak yetersiz kalmaktadır. Yirminci yüzyılının ikinci yarısında, teknolojik gelişmeler ile beraber depresyonun patogenezinde biyolojik faktörlerin rolüyle ilgili çalışmalar hız kazanmış ve psikofamakoterapide yeni çığırların açılmasına neden olmuştur. Depresyonun biyolojik etyolojisini açıklamaya yönelik geliştirilen kuramlardan, monoamin hipotezi, nörotransmitter-reseptör hipotezi ve psikonöroendokrinolojik yaklaşımlara son yıllarda nöroplastisite hipotezi eklenmiştir. Birçok postmortem ve nörolojik görüntüleme çalışmaları ile beynin yapısal durumu incelenmiş, böylece belli beyin bölgelerinin depresyonun patogenezinde rol oynadığı gösterilmiştir. Bu gözden geçirme yazımızda, depresyonun etyolojisi ve fizyopatolojisine yönelik oluşturulan kuramlar ve hipotezler gözden geçirilmiştir.

Although with continuing increases in the studies directed towards to illuminate the etiology of depression, the data we have today is not enough due to the clinical and etiological heterogenity of this mental ilness. In the second half of the twenty first century, with technological advances, the studies related to biological factors in the pathogenesis of depression has been increased and it has been caused to mark a new era in psychopharmacotherapy. In recent [year]s, neuroplasticity hypothesis was added to the theories which developed to explain the biological etiology of depression such as monoamine hypothesis, neurotransmitter-receptor hypothesis and the psychoneuroendocrinologic approaches. The structural status of the brain was evaluated by many postmortem and neurological imaging studies, thus shown certain brain regions play a role in the pathogenesis of depression. In this review article, the theories and hypotheses generated based on the pathophysiology and etiology of depression is reviewed.
