Amaç: Serotonin (5-HT) merkezi sinir sisteminde anahtar role sahip bir nörotransmitterdir. Serotonerjik disfonksi-yonun şizofreni patogenezinde etkili olabileceği bildirilmektedir. Serotonin (5-hidroksitriptamin, 5-HT2A) reseptörü insanda 13q14-21’de yerleşmiş bir gen tarafından kodlanır. Bu çalışmada 5-HT2A reseptörü geni promotör bölge-sindeki -1438 G/A polimorfizmi ile şizofreni arasında bir ilişkinin var olup olmadığı değerlendirilmiştir. Yöntem: Çalışmamıza DSM-IV psikiyatrik tanı sınıflamasına göre şizofreni tanısı konulan 115 hasta ve kontrol grubu olarak da 86 sağlıklı birey alındı. Serotonin reseptörü -1438 G/A polimorfizmi PCR-RFLP yöntemiyle, genotipler ise agaroz jel elektroforezi ve jel görüntüleme sistemi kullanılarak belirlendi. İstatistiksel analiz için SPSS 12.0 paket programı kullanıldı. Sonuçlar: Şizofreni hastalarında AA genotipi oranı %27, GA genotipi oranı %51 ve GG geno-tipi oranı %22; kontrollerde AA genotipi oranı %17, GA genotipi oranı %56 ve GG genotipi oranı %27 bulundu. AA, AG ve GG genotipleri açısından şizofreni hastaları kontrol grubu ile karşılaştırıldığında anlamlı bir fark gözlenmedi. AA genotipi taşıyan bireylerin şizofreni riski, AG ve GG genotipi taşıyanlara göre, sırasıyla, 1.13 ve 1.90 daha fazladır. Bununla beraber, fark istatistiksel olarak anlamlı değildir. Şizofreni hastalarında ve sağlıklı bireylerde 5-HT2A reseptörü A ve G alel sıklığı sırasıyla, 0.53/0.47 ve 0.45/0.55 olarak hesaplandı. Tartışma: Bulgularımız 5-HT2A reseptörü geni polimorfizmi ile şizofreni arasında bir ilişki bulunmayabileceğini göstermek-tedir.

Objective: Serotonin (5-HT) plays a key role in central nervous system. Serotonergic dysfunction may be involved in the pathogenesis of schizophrenia. 5-HT2A receptor gene maps to 13q14-21. This study aimed to investigate possible association between -1438 G/A polymorphism and schizophrenia. Methods: A hundred and fifteen schizophrenic patients diagnosed according to DSM-IV and 86 healthy control subjects were included in this study. The -1438 G/A polymorphism of serotonin receptor gene were determined by PCR-RFLP assay, genotypes were determined using agarose gel electrophoresis and image analyzer system. Statistical analyses were performed using SPSS for Windows version 12.0. Results: The frequencies AA, AG ad GG genotypes were %27, %51 and %22 in schizophrenia patients; %17, %56 and %27 in controls. The differences were not signify-cant when compared with AA, Ag and GG genotypes. Individuals with AA genotype had nearly 1.13-fold and 1.90-fold increased risk for schizophrenia than individuals carrying genotypes AG and GG, respectively. However, the differences were not statistically significant. The frequencies of A and G alleles were 0.53 and 0.47 in patients and 0.45 and 0.55 in controls, respectively. Conclusion: These findings indicate that there may not be any asso-ciation between 5-HT2A receptor gene polymorphism and schizophrenia.
