Kimyasal yapısı klozapine çok benzeyen olanzapin, güçlü etkili yeni atipik antipsikotik ajandır.Thienobenzodiazepin grubundan olan olanzapinin D2 reseptörüne göre 5-HT2 reseptörüne bağlanma afinitesi daha fazladır ve mezolimbik seçiciliği vardır.Çalışma sonuçları, olanzapinin şizofreninin pozitif semptomları üzerine haloperidol kadar etkili olduğunu, negatif semptomların tedavisinde ise haloperidolden daha etkili olduğunu göstermektedir.Hematolojik yan etkisi gözlenmeyen ve haloperidole göre EPS riski düşük olan olanzapinle genel klinik tecrübeler, iyi tolere edildiği şeklindedir.Bu yazıda, olanzapinin farmakolojik özellikleri, klinik etkinliği ve yan etkileri kısaca gözden geçirilmiştir.

Olanzapine is a new "atypical" antipsychotic agent that has a similar chemical structure with clozapine.Olanzapine, which belongs to the thienobenzodiazepine class, has a greater binding affinity for 5-HT2 compared to D2 receptors and has a mesolimbic selectivity.Results shows that olanzapine is as effective as haloperidol for positive symptoms and more effective than haloperidol for the treatment of the negative symptoms of schizophrenia.The overall clinical experience with olanzapin, which has no hematolojic side effect and low risk for EPS than haloperidol, has been one of high tolerability.In this article, the pharmacological properties, clinical efficacy and side effects of olanzapine is briefly reviewed.
