Major Depresif Bozukluk en sık rastlanan psikiyatrik hastalıklardan biridir. Patogenezini açıklamak amacı ile şimdiye kadar yapılmış birçok çalışma olsa da henüz bu konuda kesin bilgilere sahip değiliz. Ancak çalışmalar etiyolojide önemli role sahip etmenlerden birinin de genetik yatkınlık olduğuna işaret etmektedir. Çok sayıda genin depresyon etiyolojis-inde rol oynadığıdüşünülmektedir ama bu genlerin büyük çoğunluğu serotonerjik ve glukokortikoid sistemi ile ilişkilidir. Bahsedilen bu genetik varyantların beyinde yapısal ve fonksiyonel değişikliklere yol açarak depresyona zemin hazırladığı düşünülmektedir. Bunların dışında son zamanlarda genetik çalışmalar gen-çevre etkileşimi ve endofenotip kavramına da yoğunlaşmıştır.

Major depressive disorder is one of the most common psychiatric diseases. Despite the great effort to find the pathogenesis of it, we fairly know about how MDD develops. Among the etiological factors, hereditability of the disease is the best known. Although different candidate genes were proposed by different investigations, the most consisted ones were related to serotonin and glucorticoid system. These genes might affect the functional and structural status of the depressed brain. Current approaches corroborate phenomena related to endophenotype and gene-environment interactions.
