Obsesif-kompulsif bozuklukta genetik çalışmalar

Obsesif-kompulsif bozukluk (OKB) zorla gelen yineleyici fikir, hayal ve dürtüler yani obsesyonlar ve katı, törensel ve zaman kaybına yol açan davranışlar, yani kompulsiyonlardan oluşan bir psikiyatrik bozukluktur. OKB’nin kalıtım özelliğine ilişkin ilk bulgular aile ve ikiz çalışmaları ndan elde edilmiştir. Segregasyon analizleri ise OKB kalıtımında majör bir gen bölgesinin sorumlu olabileceğine ilişkin bilgiler sağlamıştır. Yayımlanmış bulunan tek bağlantı çalışmasında 9p kromozom üzerinde çok noktalı bir bağlantı bulunduğu bildirilmiştir. Son dönemde ise genetik çalışmalar OKB etiyolojisinde rolü olduğu düşünülen bazı aday genler üzerinde yoğunlaşmış, özellikle serotonerjik ve dopaminerjik sisteme ilişkin genler üzerinde yapılan çalışmalar bu sistemlere ilişkin önceki klinik bulguları destekleyen sonuçlar sağlamıştır. Ayrıca, opioid, glutamaterjik ve immun sistemlere ilişkin genler ile nörogeli- şimsel genlere ilişkin ilk çalışma sonuçları gelecekteki çalışmalar açı- sından araştırmacılara umut veren genler olarak belirlenmiştir. Bu yazıda OKB’ye ilişkin aile, ikiz, segregasyon, bağlantı ve aday gen çalı şmaları gözden geçirilmektedir.(
Genetic findings of obsessive-compulsive disorder
Obsessive-compulsive disorder (OCD) is characterized by repetitive, intrusive ideations, image or urges (obsession) and/or rigid, ritualistic and time-consuming behaviours (compulsions). Early evidences on inheritance of OCD have been obtained from family and twin studies. Segregation analysis provides basic support for existence of a major gene locus. In only published linkage analysis, it was reported that multipoint linkage was found on chromosome 9p. Recently, genetic studies have centered on plausible candidate genes, which are thought to be involved in the etiology of OCD. Preliminary findings from the studies of several candidate genes have contributed to previous clinical findings, especially related to serotonergic and dopaminergic systems. Also, the candidate genes of opioid, glutamatergic systems as well as the genes of immune system and neurodevelopmental progress give hope to researchers for future investigation of these systems. In this review, the family, twin, segregation, linkage, and candidate genes studies of OCD will be reviewed
