Hipotalamustaki nörosekretuvar nöronlar, salgıladıkları salgılatıcı (releasing) ve engelleyici (inhibiting) hormon veya faktörler aracılığıyla ön hipofiz hormonlarının salgılanmasını kontrol ederler. Karşılıklı etkileşim halinde olan hipotalamus ve ön hipofiz hormonları, vücuttaki ana hormon eksenlerinin düzenlendiği bir üst merkez konumundadır. Bu hormon eksenleri hipotalamo-hipofizer-adrenal eksen, hipotalamo-hipofizer-tiroid ekseni, hipotalamo-hipofizer-gonad eksen, büyüme hormonu ve prolaktin eksenleridir. Antidiüretik hormon (ADH) ve oksitosin ise hipotalamusun paraventriküler ve supraoptik nukleuslarındaki nöronların aksonal uzantıları ile arka hipofize (nörohipofiz) ulaşarak burada depolanır ve sistemik dolaşıma salınırlar. Ön hipofiz hormonlarının klasik fizyolojik etkileri dışında davranış üzerine de çeşitli etkileri vardır. Oksitosin ve ADH'nin ise bazı toplumsal kaynaklı olmayan (içgüdüsel) davranışlar üzerinde etkisi bulunmaktadır. Bu derlemede hipotalamo-hipofizer eksenlerin klinik fizyolojisi ana hatlarıyla özetlenecektir.

Neurosecretory neurons of hypothalamus control the secretion of anterior pituitary hormones via their releasing and inhibiting hormones or factors. Interacting hypothalamic and pituitary hormones are the upper center that regulates the main hormonal axes of the body. These hormonal axes are hypothalamic-pituitary-adrenal axis, hypothalamic-pituitary-thyroid axis, hypothalamic-pituitary-gonadal axis, growth hormone axis, and prolactin axis. Antidiuretic hormone (ADH) and oxytocin release down the axons of the supraoptic and paraventricular nuclei of the hypothalamus into the posterior pituitary, where they are stored and released into the systemic circulation. Anterior pituitary hormones have various behavioral effects other than the classical physiological effects. Oxytocin and ADH have effects on some non community-acquired (instinctive) behaviors. The clinical physiology of the hpothalamic-pituitary axes will be summarized in this review.
