Amaç: Bu çalışmada otistik bozukluğu olan çocukların plazmalarında glutatyon peroksidaz (GSH-Px), selenyum (Se), çinko (Zn), mangan (Mn), bakır (Cu); eritrositlerinde süperoksid dismutaz (SOD), GSH-Px, Se, Zn, Mn düzeylerinin araştırılması ve normal çocuklarla karşılaştırılması amaçlandı. Yöntem: Araştırma grubunu, DSM-IV tanı ölçütlerine göre otistik bozukluk tanısı konan 4-12 yaş grubu 45 çocuk (39 erkek, 6 kız) oluşturdu. Kontrol grubu olarak da herhangi bir tıbbi hastalığı olmayan 4-12 yaş grubu 41 çocuk (35 erkek, 6 kız) seçildi. Eritrositlerde SOD, eritrosit ve plazmada GSH-Px ölçümleri spektrofotometrik olarak; eritrositlerdeki ve plazmadaki bakır, çinko, selenyum ve mangan ölçümleri ise atomik absorbsiyon spektrofotometresi kullanılarak yapıldı. Sonuç: Araştırma grubundaki otistik bozukluğu olan çocuklarda normal çocuklara göre eritrosit SOD ile eritrosit ve plazma GSH-Px etkinliğinde, eritrosit ve plazma selenyum, çinko ve mangan düzeylerinde istatistiksel olarak anlamlı bir azalmanın olduğu bulundu. Otistik bozukluğu olan çocukların plazma bakır düzeylerinde ise kontrol grubundaki normal çocuklara göre anlamlı bir artışın olduğu saptandı. Tartışma: Bu sonuçlar otistik bozuklukta, normal çocuklara göre antioksidan enzim savunma sistemi işlevlerinde azalmanın olduğunu ve bu nedenle biriken serbest radikallere bağlı olarak beyin dokusunda örselenmenin olabileceğini düşündürmektedir. Otistik çocuklarda plazmadaki ve eritrositlerdeki düşük düzeydeki eser elementlerin SOD ve GSH-Px işlevlerinde azalmaya neden olduğu ileri sürülebilir. Yaptığımız çalışmada söz konusu olan bu durum, plazma bakırı için geçerli bulunmamıştır.

Objective: The plasma levels of glutathion peroxidase (GSH-Px), Se, Zn, Mn, Cu and superoxide dismutase (SOD), GSH-Px, Se, Zn, Mn in the erythrocyte were investigated in the children with autistic disorder and compared with normals in this study. Method: The study group were consisted of 45 children (39 boys, 6 girls), age range between 4 and 12 [year]s, and met DSM IV criteria for autistic disorder. The control group were consisted of 41 children (35 boys, 6 girls), age range between 4 and 12 [year]s, and had no history of any medical diseases and psychiatric disorders. Levels of erytrocyte SOD, erythrocyte and plasma GSH-Px were performed spectrophotometrically, erythrocyte and plasma zinc, selenium, manganese, and plasma copper were measured with atomic absobtion spectrophotometry. Results: Activity of erytrocyte SOD, erythrocyte and plasma GSH-Px, erythrocyte and plasma selenium, zinc and manganese levels in the cihldren with autistic disorder were significiantly lower than normals. Plasma copper levels of children with autistic disorder were significiantly higher than normals. Conclusion: These results assumed that children with autism have low levels of activity of antioxidant enzyme systems, for that reason the accumulation of the free radicals could damage brain tissue. It was suggested that low levels of plasma and erythrocyte trace elements may cause low activity levels of SOD and GSH-Px in the children with autistic disorder. This suggestion was not valid for plasma copper.
