Klozapin, klasik antipsikotiklere dirençli veya bu tür ilaçları yan etki nedeniyle alamayan şizofrenik hastaların önemli bir kısmında (%30-50) etkilidir. Klinik çalışmalarda, klozapinin yan etkileri düşük olmakla birlikte şizofreninin negatif ve pozitif belirtilerini bastırmada etkili olduğu gösterilmiştir. Tedavi edici etkisine karşın, agranülositoz sıklığı (hastaların %1-2’sinde) diğer antipsikotiklerden göreceli olarak daha yüksektir. Epileptik nöbetler klozapin tedavisinin önemli bir yan etkisidir. Nöbet riski titrasyonun hızlı yükselmesine ve yüksek doza bağlı bir etki olduğunu açıkça doğrulamamıştır. Bu yazıda, klozapin tedavisi sırasında nöbet görülen iki olgu sunulmuş ve tartışılmıştır.

Clozapine is effective in a substantial proportion (30 to 50%) of schizophrenic patients who are refractory to or intolerant of classical antipsychotic drugs. Clinical studies have shown clozapine to be effective in suppressing both the positive and negative symptoms of schizophrenia and to be associated with an extremely low incidence of side effects. Despite its promising therapeutic potential, the incidence of clozapine- induced agranulcytosis (1-2% of patients) is relatively higher than other antipsychotics. Seizures are an important side effect of clozapine therapy. The risk of seizures may be increased by rapid upward titration and higher doses. Recent data, however, do not clearly confirm the dose-dependent effect. In this report, two cases with seizures during clozapine therapy are presented and discussed.
