Uyku Bozukluğu Genetiği

Uyku alanındaki genetik çalışmalar uyku bozukluklarının oluş mekanizmalarını DNA düzeyinde açıklamaya çalışmaktadır. Uykunun genetik olarak kontrol edildiği bilinmektedir fakat tespit edilmiş tek bir uyku geni bulunmamaktadır. Uyku bozuklukları birçok genle ilişkili ve çevresel etkileşimin daha sık olduğu kompleks kalıtım özellikleri göstermektedir. Genetik ilişkisi çok sayıda çalışmalarla doğrulanan hastalıklar obstrüktif uyku apne sendromu, narkolepsi ve huzursuz bacaklar sendromudur. Obstrüktif uyku apne sendromunun apolipoprotein E4, tümör nekrozis faktör ve anjiotensin dönüştürücü enzim kodlayan genlerle ilişkili bulunmuştur. Narkolepsi ile HLA, CPT1B, CHKB, T hücre reseptörü α loküsü, P2RY11, DNMT1, CTHS, TNFSF4 ilişkisi ve huzursuz bacaklar sendromu ile BTBD9 (6p), MEIS1 (2p), MAP2K5/LBXCOR1 (15q), PTPRD (9p), nNOS (12q) genleri ilişkisi de bulunan diğer önemli sonuçlardır. Uyku bozukluklarının genetik temellerinin
tespiti hastalık nedenlerinin ve oluş mekanizmalarının aydınlatılmasını sağlamanın yanı sıra tanı ve tedavide de yeni ufuklar açabilir.
Genetics of Sleep Disorders
Genetic studies about sleep attempts to explain mechanisms of sleep disorders at the level of DNA. It is known that sleep is genetically controlled, but there is no single gene has been identified. Sleep disorders are complex genetic disorders which are associated with multiple genes and the environment. Obstructive sleep apnea syndrome, narcolepsy and restless legs syndrome are diseases which have genetic association in the literature. Genes encoding apolipoprotein E4, tumor necrosis factor and angiotensin converting enzyme are significantly associated with obstructive sleep apnea syndrome. Association between narcolepsy and HLA, CPT1B, CHKB, T-cell receptor a locus, P2RY11, DNMT1, CTHS, TNFSF4 and between restless legs syndrome and BTBD9 (6p), MEIS1 (2p), MAP2K5/LBXCOR1 (15q), PTPRD (9p) and nNOS (12q) are the other significant results. Identification of the genetic basis of sleep disorders can create new horizons for the diagnosis and treatment as well as clarification of the causes and mechanism of diseases.
